Establishment of AFLP Analysis System and its Application in Chaenomeles

Twenty-nine cultivars in the genus Chaenomeles were used as materials to establish the AFLP(Amplified Fragment Length Polymorphism)reaction system for studying the relationship and genetic diversity in flowering quince resources.The AFLP analysis system was established by detected the results of several reactions such as DNA extraction,enzymes restriction,pre-amplification and selective amplification;the main factors which affected AFLP analysis were analyzed.Eight pairs of primers,E-AAG+M-CAA,E-ACA+M-CAA,E-AAG+M-CAC,E-AAG+M-CAG,E-AAG+M-CAT,E-AAG+M-CTG,E-ACA+M-CTT,and E-ACG+M-CTT,were selected to selectively amplify the genomic DNA of the cultivars,and clear amplification bands were obtained.The results suggested that the reaction system was good for Chaenomeles AFLP analysis.
