The Relationship Between Survivin Gene and Protein Expression and Proliferation and Apoptosis of Human Clear-cell Renal Cell Carcinoma 786-0

Objective To study the correlation between Survivin gene and protein expression and proliferation and apoptosis of human 786-0 renal clear cancer cells. Methods Human renal cell carcinoma cell line 786-0 was used for survivin transfection group, while renal tubular epithelial cell HK-2 was used as negative control group and transfected with survivin mimic. The expression levels of survivin gene and protein was determined72 h after transfection. Absorbance value, cell number and ratio of apoptotic cells in two groups were compared; Pearson correlation analysis was conducted based on Survivin gene and protein expression levels and cell light absorbance value, cell number and apoptotic cell ratio. Results The expressions of Survivin mRNA and Survivin protein in Survivin transfection group were significantly higher than that in negative control group(P 0. 05). The absorbance value of Survivin transfection group was significantly lower than that in negative control group( P 0. 05), From the 2 nd day, the number of cells in transfection group was significantly lower than the negative control group(P 0.05). The percentage of apoptosis in 786-0 cells was significantly lower than that in the negative control group from the first day(P 0. 05). Pearson correlation analysis showed that the expression levels of Survivin gene and protein were inversely proportional to the absorbance value and cell number,and were positively proportional to the ratio of apoptotic cells. Conclusion Survivin gene and protein expressions can inhibit the proliferation of 786-0 renal clear cancer cells and promote apoptosis.
