Study on Immunogold Quantitative Assay for Microalbuminuria

Using gold sol particles as the carrier of immuno-ligands,an immunogold agglutination inhibition assay was establishead to quantify microalbuminuria.In this assay system,the suitable diameters of the gold sol particles were 5070nm.The flocculated sediment of the collidal gold in urine medium was diminished by using borate buffer as the reaction buffer.A final concerntration of 3% of polyethylene glycol 6000 ensured the agglutination test to be fulfilled completely.The linear range of the calibration curve of the method was 40200mg/L,which was suitable for the assay of microalbuminuria.103 clinical urine specimens were determined by this method,and their results were compared with those of rate nephelometry on automated protein analyzer.The correlation of these two methods reached 995%.
