An Effective Method for Isolation of High-quality Total RNA from Fruit Pulps

Although isolation of total plant RNA has become a conventional technique,owing to the particularity of fruit material,there is not yet ideal method for the isolation of total RNA.Based on previous CTAB method,an common method for isolation of total RNA from fruit was discussed in order to lay the foundation on fruit development molecular biology.Total RNA was successfully isolated from fresh apple,peach,strawberry and grape fruits by the effective CTAB method modified with a 16% of β-Mercatoethanol and 3% of PVP in extraction buffer,1/3(V/V)of 5 mol/L KAC(pH4.8)in supernatant.The isolated RNA was assayed by ultraviolet spectrophotometry,electrophoresis,reverse transcription and RT-PCR and showed that the modified CTAB protocol was preferred method for isolation of high-quality total RNA from fruit pulp.Soybean pollen-tube.
