Establishment of liquid culture system for chestnut(Castanea mollissima Blume.)pollen

【Objective】To optimize the liquid culture system of Chinese chestnut(Castanea mollissima Blume.)(‘Yanhong')pollen,we investigated the effects of the concentration of sucrose,H3BO3,CaCl2 and other factors on pollen germination rate.【Methods】The single factor such as sucrose,H3BO3 and CaCl2was tested.Based on the above result,orthogonal experiment was carried out to optimize the liquid culture condition for Chinese chestnut pollen.【Results】The results showed that the medium contained 15% sucrose,0.005% H3BO3,0.04% CaCl2,pH 6.5,34℃ and 175r/min were optimal for the pollen germination of Chinese chestnut.Under the above cultural condition,the pollen germination rate reached 28.86%,even the pollens had been preserved for one year.【Conclusion】The results indicated that pollen germination rates were mainly affected by several factors such as medium components,pH value,rotation speed and culture temperature.
