Effects of ginsenosides on myocardial ischemia and reperfusioninjury and concentrationeffect relationship

Objective:To study the effects of ginsenosides (GS) on myocardial ischemia and reperfusion injury,and concentrationeffect relationship.Methods:①Eleven mongral dogs were divided into 2 groups randomly and cardiopulmonary bypass was estabilished.In GS group,20 mg/L of GS was added into the cardioplegia.The indexes of myocardial dynamics and hemodynamics were recorded at 15 min after aortic unclamping.The samples were taken from the right ventricle and mitochodria were scored under electron microscope at 60 min of aortic crossclamp and 15 min after unclamping of aorta.②Thirty rabbits were divided into 5 groups randomly,and then isolated working hearts underwent 60 min normothermic ischemia by aortic crossclamping.In GS group 20,40,80,160 mg/L of GS were added into the cardioplegia respectively..As mentioned above,the parameters (at 25 min of aortic unclamping) and the myocardial samples were obtained.In addition the contents of calcium in the cardium were measured.Results:Before and after the dogs′cardiopulmonary bypass and the isolated rabbit hearts′working were established,the data of myocardial dynamics (±dp/dtmax),hemodynamics (CI),mitochodria score and the myocardial calcium content in GS group were better than those in control group (P0001),and those in GS 80 mg/L group better than those in 20 and 40 mg/L groups,whereas all the data mentioned above in GS 160 mg/L group were lower than those in control gorup.Conclusion:①GS 20 ̄80 mg/L has significantly protective effect on the myocardial ischemia and reperfusion injury,and the effects increase when the concentration of GS increases;whereas the effects are converse when the concentration of GS was 160 mg/L.②GS has the calcium channel block effect.
