PURIFICATION AND PROPERTIES OF CELLULASE FROM ASPERGILLUS NIGER VAR

One β- glucosidase and be endoglucanases from Aspergillus niger var were purified to homogeneous by (NH4)2SO4 precipitation, Sephadex G- 100 Chromatography,DEAE-Sephadex A50 chromatography and HA chromatography. They were designated to be βG, EGⅠ, EGⅡ, EGⅢ. The moleculur weight of the four Components were estimated to be 1.062 × 105, 2.51× 104, 2.95 ×104, 2.25 × 104 respectively. The optimum pH of enzyme were 6.0,5.5, 5.5, 5.0, and the optimum temperature of the enzyme were 50, 45, 45, 50℃ accordingly.Enzyme activity was inhibital strongly by Ag+ and Hg2+; but slightly activated by Co2+, Ni2+ and Mn2+.
