DETECTION OF AUTOCRINE GROWTH FACTORS IN THE SERUM-FREE CULTURED INTESTINAL CARCINOMA CELL LINE HICSF

A comparative study was conducted on the expression and response of some growth factors and cytokines between the serum cultured (HIC) and the sertm-free cultured (HICSF) intestinal carcinoma cells in order to investigate the autocrine regulation mechanism of HICSF cell line. A RT- PCR technique was utilized to detect the expression of bFGF, TGFβ1,IL- 6, IL- 8, G- CSF, IL- 6R and IL-8R between the tWo Cells, and a 3H- TdR incorporation method was conducred on the observation of the effect of recombinant IL-6 and TGFβ1 on the proliferation of both cell lines. It was found that IL-6, IL-6R and G- CSF were expressed at higher levels in HICSF cells than in HIC cells, and that the expression of IL-8, IL-8R and TGFβ1 was lower in HICSF cells than that in HIC cells, and no significant difference was found in the expression of bFGF between both cells. Recombinant IL-6 and TGFβ1 stimulated the prolifhation of both cells, the stimulatory effect of IL-6 and TGFβ1 on HICSF cells was markedly higher than that on HIC cells. These results suggested that bFGF, TGFβ1, IL- 6, IL- 8 and G- CSF may be involved in the autocrine regulation of the proliferation of intestinal carcinoma cell, and that IL- 6, TGFβ1 and G- CSF may play an important role in the autocrine growth of HICSF cell in serum- free medium.
