THE EFFECT OF TRYPSIN TREATMENT ON PLASMA MEMBRANE H~+-ATPase FROM SOYBEAN HYPOCOTYLS

The effect of trypsin treatment on plasma membrane H- + -A TPase is studied using plasma membrane vesicles purified from soybean hypocotyls by sucrose gradient centrifugation method. Results show that under mild trypsin treatment the ATPase hydrolysis activity increases and optimum pH value moves f rom 6.5 to 7.0. PNPP hydrolysis activity is stimulated as well under trypsin tre atment. In the presence of 10~30 mmol·L- -1 PNPP, the PNPP hydroly sis activity of the cleaved ATPase increases by 21%~28%. Kinetic studies ind icate that the K m values for PNPP hydrolysis is reduced from 2.66 to 1 .44 mmol·L- -1 by proteolysis. Taken together, the structure and a ctivity of the phosphatase domain are affected by trypsin treatment, implying th at this domain might be regulated by the C-terminal end of plasma membrane H- +-ATPase.
