CALLUS INDUCTION AND ROOT ORGANOGENESIS FROM LEAVES OF ZEA MAYS SEEDLINGS

Leaf segments from maize seedlings about 7 days old are used as materials in the experiment. After seedlings are pretreated to growing on the 1/2MS basic medium supplementary with 1 mg·L -1 KT or 1 mg·L -1 BA, callus induct rate from basic segment of 2nd leaf (numbered from outside to inner) is higher (up to 58.3% ) than that from first one on the MS medium added with 2 mg·L -1 2,4-D, and adventitious roots are regenerated at different rates after two-week culture. When 2,4-D and 1 mg·L -1 BA or 1 mg·L -1 KT are combined to induce the callus, the induction of callus and adventitious roots is inhibited sharply. However, when 500 mg·L -1 CH is added into the callus induction medium with or without cytokinin, the rate of callus and root formation is increased with comparsion to the counterparts.
