Effects of LDL on Movement Characteristic of Bovine Frozen Sperm

In order to protect the spermatozoa against cold shock,hen egg yolk is widely used as a cryoprotective agent in semen freezing extenders.The protective action of yolk is largely presumed to be due to low density lipoprotein(LDL).The objective of the present study was to obtain information about the cryoprotective effect of different concentrations LDL(7%,8%,9%,10%,w/v) in bovine semen extender instead of egg yolk in 0.25 ml straws.The characteristics of sperm motion after frozen-thawed were evaluated with a computer-aided sperm analysis(CASA) system(Weili Software Co.Ltd,Beijing,China).The results showed that the sperm motion characteristics were improved with LDL in the extender,as compared to the extender containing 20% yolk.The highest sperm motility was obtained with 8% LDL(w/v) and it was 55.6%,which was significantly higher than that of other groups(P 0.01).The straight line velocity reached 32.5μm/s,the velocity of the average path was about 35.7μ m/s,and they were significantly higher with semen frozen in 8% LDL extender compared to control group(P 0.05).The percentage of the sperm of grade a and b was 50%.This study affords some way to the exploitation and utilize of bovine frozen sperm.
