Therapeutic effect of previously activated psoralens on transplanted leukemic tumour in nude mice

Objective To investigate the anti-leukemic effect of previously activated psoralens(PAP) in vivo in mice.Methods HL-60 ceils were transplanted subcutaneously on the back of nude mice.Experi- mental group were injected PAP for several successive days on various sites since different time points.Con- trol group were injected normal saline correspondingly.Differences in tumour load,turnout forming rate and overall survival between two groups were compared.Results At 8th day after transplantation of HL-60 cells tumour-forming-rate(TFR)was 100 %.Intra-tumour injection trial revealed that mean values of tumour load in PAP and control arm are(3.21±1.32)g and(5.0±1.0)g respectively,the difference in tumour load between two arms was significant statistically(t=3.42,P=0.0031).The average survival times(AST)in PAP and con- trol arm are(35±9.92)d and(28.3±5.14)d respectively,the difference in AST between two arms was not sig- nificant(Z=1.52,P=0.1294).Pathological examination discovered that necrotic field of tumour in experi- mental group was obviously larger than that in control group.Intramuscular injection trial revealed that the TFR in experimental and control arm are 80 % and 100 % respectively,the difference in TFR between two arms was not significant(x~2=2.22,P=0.1360).The ASTs in PAP and control arm are(31.5±6.39)d and (17.8±12.07)d respectively,the difference in AST between two arms was not significant statistically(Z=1.37, P=0.1713).Transplanting site injection trial showed that the TFR in PAP and control arm are 60 % and 100 % respectively,the difference in TFR between two arms was different significanfly(x~2=5.0,P=0.0250), the AST in PAP and control arm are(37.9±6.21)d and(20.8±4.10)d respectively,the difference in AST be- tween two areas was also different significantly(Z=4.30,P=0.0000).Condusion PAP could kill transplant- ed leukemic cells in vivo in nude mice.
