Experimental Study on Effects of Total Glucosde Extracted from Cynanchus Auriculatus (CA) in Inhibiting Tumors

Cynanchus auriculatus is a valuable tonic. The experiment studied the effect of total glucoside extracted from it (CA) in inhibiting tumors in vivo and vitro. After hypodermic transplantation of 3×10~5 Lewis lung carcinoma cells, C57BL/6 inbred male mice were divided into CA group (CA 225mg/kg×10), CY group (CY 40mg/kg×2), CA + CY group (CA150mg/kg×10 +CY40mg/kg×2)and control group(DWoo. 2ml/miase×10). All animals were given drugs peritoneally. The measurement of tumor weight, lung metastasis and FCM assay were performed in vivo. The cytotoxicity test, ~3HTdR incorperation test and morphological observation were used for examining the effects of CA on the cultural cells of EAC and S—180 tumors. The results showed that the tumor weight in the CA group (1.36g )and CA+CY group (0.52g) was lighter than that of the control (2.21g) and CY group ( 0.75g ) respectively. The tumor inhibition rate of the CA and CA+CY groups reached 38.68% and 76.27%. The extent of lung metastasis in these two groups was also less severe. It was indicated by flow cytometry that in the CA group the number of Lewis luong carcinoma cells in G1 phase increased and number of cells in Sphase reduced significantly as compared with the control(p0.001 and p0.05). The number of EAC and S—180 living cells cultured in different CA doses all reduced. The growth inhibition rate of 2mg CA dose for EAC and S—180 cells reached 73.6% and 43.1% respectively. Under the effects of CA, morphological changes of these two cell clones such as cellular swelling, membrane rupfure and chromatin changes were observed. It was also confirmed that CA can inhibit ~3HTdR incorperation into DNA of EAC and S—180 tumor cells.
