Study on extraction conditions optimization of total flavonoids from Ophiopogon japonicus by response surface methodology and its antioxidant effects

Objective:To investigate the extraction technology and antioxidant effects oftotal flavonoids from Ophiopogon japonicus(TFOJ).Methods:The extraction conditions optimized by response surface methodology.The free radicals scavenging activity ofTFOJ, TFOJE(ethyl acetate fraction ofTFOJ) and TFOJB(n-butanol ofTFOJ) was measured in vitro.The protective effects ofTFOJE and TFOJB on D-galactose induced oxidative damage mice were analyzed in vivo.Results:It obtained 8.86 mg flavonoids from 1g dry powder ofOphiopogon japonicus on the optimum conditions:extraction time was 188 min, ethanol concentration was 73%, temperature was 72 ℃, and feed/solvent ratio was 1:29.TFOJ, TFOJE and TFOJB showed effective activity on scavenging DPPH and ABTS+ free radicals.TFOJ-B had more effective antioxidant effects than TFOJE, and it might decrease the MDA content and increase the GSH-Px, SOD activities, the decrease rate ofMDA contents in serum and liver were 17.2 % and 15.7 %, the increase rate ofGSH-Px activities in serum and liver were 36.5 % and 34.2 %, the increase rate ofSOD activities in serum and liver were 17.2 % and 13.5 %.Conclusion:The flavonoids from Ophiopogon japonicus had obvious free radicals scavenging effects, and its might reverse oxidative damage ofmice.
