In vitro culture of Ri-plasmid transformed root of Cassia obtusifolia L.

To transform Cassia obtusifolia L. using Ri plasmid and determine the production of anthraquinones in the cultures of transformed roots. Methods: Ri plasmid transformed roots from explants of Cassia obtusifolia L. were induced by leaf-disc technique and identified by Southern hybridization. The growth period was studied and the contents of six free anthraquinones of the roots quantitatively determined by high performance liquid chromatography (HPLC) technique. Results: Results of Southern hybridization indicated that T-DNA of Ri plasmid was transferred into genome of the transformed roots. HPLC results showed that the total amount of six anthrequinones of the roots cultured under light condition (2000 lx, 24 h/d) was twice higher than that under dark condition (24 h/d). Conclusion: The culture period of 35 d was suitables for the transformed root, and dark condition was found to be favorable for growth of the roots and accumulation Of the free anthraquinones.
