The changes in pulmonary vascular structure and plasma level of nitric oxide in hypoxic pulmonary hypertensive rats

Objective: To explore the possible role of nitric oxide (NO) in the development of hypoxic pulmonary hypertension. Methods: A total of 12 Wistar rats (6 pairs ) were involved in the whole process of the experiment. Pulmonary artery pressure of each rat was evaluated by using a right cardiac catheterization procedure. Plasma concentration of NO was measured indirecthy via spectrophotometry. Changes in pulmonary vascular structure were measured and the ultrastructural changes in intrapulmonary muscularized arteries were observed. Results: Pulmonary artery mean pressure was significantly increased in rats exposed to two week hypoxia [(2.75±0.30) kPa vs ( 2.06 ±0.14) kPa, P 0.05]. Meanwhile, indirect plasma NO concentration was markedly decreased in comparison with controls[(3.5±1.2) μmol·L -1 vs (4.8±0.4) μmol·L -1 , P 0.05]. Relative medial thickness (RMT) and relative medial area (RMA) of pulmonary arteries and the percentage of muscularized arteries of small pulmonary vessels were obviously increased in hypoxic rats as compared with those of normal controls [RMT of median pulmonary arteries: (9.94±0.30)% vs ( 7.70 ± 0.41 )%; RMA of median pulmonary arteries: (18.87±0.54)% vs (14.76±0.75)%; RMT of small pulmonary arteries: ( 15.15 ±0.21)% vs (10.12±0.60)%; RMA of small pulmonary arteries: ( 27.99 ±0.88)% vs (19.23±1.05)%, P 0.01, respectively]. Ultrastructural changes, such as hyperplasia, hypertrophy and swelling of endothelial cells and hyperplasia, hypertrophy and the increased number of synthetic phenotype of smooth muscle cells, were found in intrapulmonary muscularized arteries of hypoxic rats. Conclusion: The down regulation of NO due to hypoxia is probably involved in the development of hypoxic pulmonary vascular structural remodeling and even hypoxic pulmonary hypertension.
