The Studies on Technique Conditions of Protoplast Isolation and Purification of Three Species of Rhododendron

Factors affecting protoplast Isolation and purification of Rhododendron cultivars three species of R.fortunei,R.hybridum and R.pulchrum in vitro,including different combinations of enzyme mixture,dissolved time,pH value of enzyme liquid,material physiological state,centrifugal speed and time,were studied.The yield and viability of protoplast was measured,through different combination of enzyme,different dissolved times,and when the separation at low-speed of 1 000 r/min.The results showed that the optimised method for leaves in vitro from the Rhododendrons was to dissolve it 14 hours with a combination of 2%Cellulase E0240 +1% Fluka Pectinase,and pH 6.0 of enzyme liquid.A plenty of vigorous protoplasts of Rhododendron cultivars could be obtained at centrifugate 6 min with 1 000 r/min.The protoplast viability can be up to 79.27%.
