Genetic specificity in Native Medicago sativa L. cv. Longdong

For analyzing the genetic specificity of Longdong native alfalfa(Medicago sativa L.cv.Longdong) and estimating the genetic relationship between it and other alfalfa cultivars,the POD isozyme of 41 alfalfa cultivars including Longdong native cultivar were analyzed using the polylamide gel eletrophoresis technique.The results show that 22 enzyme bands in total appeared from the experimental materials with the largest number of enzymes at 9 and the least at 5.Native Longdong alfalfa had 6 enzyme bands and the one on Rf 0.26 as the specific enzyme band.Cluster analysis by transformation of the 0-1 matrix to genetic distance matrix show that Longdong native alfalfa was classified into a separated category.In conclusion,there is a big genetic difference between Longdong native alfalfa and the other 40 alfalfa cultivars,which providing a powerful support to the morphological particularity of Longdong native alfalfa.
