PROTECTION OF MK801 AND NIFEDIPINE FROM EMP-INDUCED DAMAGE IN HIPPOCAMPAL NEURONS IN VITRO

Objective:To investigate the probability of medicine protection on injury of hippocampal neurons in vitro induced by electromagnetic pulse(EMP).Methods:Firstly,optimal dose component of MK801(NMDAR antagonist) and nifedipine(L-Calcium ion channel blocker) were selected.Secondly,primary hippocampal neurons in vitro were exposured to EMP(6×10~4V/m,rise time 20ns,pulse width 30μs,2.5 pulses/min for 2 minutes) radiation after pretreated with MK801 and Nifedipine.Then the rate of proliferative neurons were detected with MTT. Results:MK801 20μM and MK801 20μM+Nifedipine 1μM were the optimal dose.The proliferative rate expressed by OD value in MK801 and MK801+Nifedipine group [(0.25±0.06) and(0.27±0.07) respectively] were higher than that in EMP control group(0.17±0.08,P0.05). Conclusions: Pretreatment with MK801 and Nifedipine can partly block EMP-induced damage in hippocampal neurons in vitro.
