Direct Electrochemistry of Horseradish Peroxidase Immobilized on Nano-Ag-Cysteamine-Modified Glassy Carbon Electrode

Cysteamine was electropolymerized at glassy carbon electrode by cyclic voltammetric method first,and then Ag nanoparticles and Horseradish peroxidase(HRP) were immobilized on the electrode successively through self-assembled technology.Then a new hydrogen peroxide biosensor(HRP/Ag/Cys/GCE) was fabricated.The fabricated process was followed by electrochemical impedance spectroscopy.The electrochemical characteristics of the biosensor were studied by cyclicvoltammetry,and chronoamperometry.Direct electrochemistry and electrocatalysis of HRP were investigated.HRP/Ag/Cys composite film exhibited a pair of stable and quasireversible cyclic voltammetric peaks for the HRP Fe(III)/HRP Fe(II) redox couple with a formal potential of about-300mV(vs.SCE) in a phosphate buffer solution(pH 7.0).The HRP/Ag/Cys/GCE sensor displayed good stability and electrocatalytic activity to the reduction of H2O2 without the aid of an electron transfer mediator,the respond time was less than 5 s.
