Comparison of Observation Techniques of Torenia(Torenia fournieri Lind.)Embryo Sac

Taking different staining methods and microscopes to study the embryo sac of torenia,the results showed that Leica DM5000B differential interference contrast(DIC) microscope and Leica DM5000B fluorescence microscope are not suitable to observe or take ultrastructure photo of embryo sac clearly,and that Leica DMRXA microscope can be merely satisfactory.But the laser scanning confocal microscope(LSCM) is the best choice.The embryo sac could be observed by ordinary microscope,but the effect would be better if it be stained. By Ehrlich’s hematoxylin staining,the embryo sac can be stained lightly.Stained by eosin Y,the inner structure could get appropriate effect by fluorescence microscope.Stained by fluorescence dye,such as DAPI,Alexa 488-phalloidin,propidium and rhodamine,the character of embryo sac structure could be observed and photo can be taken clearly by fluorescence microscope or LSCM.
