Effect of dexamethasone on eotaxin expression in lung tissue from rat model of asthma

Objective By investigating eotaxin expression in rat′s lung, to discuss the possible mechanism of dexamethasone′s therapeutic effect on asthma.Methods The rat model of asthma was established by the ovalbumin challenge method. The animal models were divided into three groups:control,asthma and dexamethasone treated. Eotaxin protein and mRNA were measured by immuneohistochemical and RT-PCR techniques respectively.Results In the asthmatic group, the protein level and the mRNA expressed of eotaxin(0.32±0.04;0.86±0.21)were significantly elevated compared to those in the control group(0.10±0.03;0.18±0.05), the dexamethasone treated group(0.12±0.02;0.21±0.10) were significantly decreased compared to the asthma group,and similar to those in the control group.Conclusions Eotaxin actively participated in the process of eosinophilic inflammation. Dexamethasone demonstrated anti-inflammatory effects by down-regulation of eotaxin.
