The pilot study on metastasis model of highly metastasic human large cell lung cancer cell lines in nude mice

Objective To establish metastasis model of highly metastasic human large cell lung cancer cell lines in nude mice so as to provide suitable model for the study of metastasis of lung cancer related molecular mechanism.Methods The highly metastasic human large cell lung cancer cell lines were inoculated into nude mice by left ventricle.Metastasic focus were observed by MRI in forming time and position.After 12 weeks,human tumor cells were recovered from metastasic focus,expanded in culture. After one to two weeks of culture, a pure population of human cancer cells was obtained, as confirmed by flow cytometry using a FITC-conjugated anti-human HLA-A, B, C antibody.In enatic cell and tumor cells of metastasic focus, the following parameters were studied:in vitro growth curve,plate efficiency,an analysis of the cell cycle.Results Metastasic focus became detectable by MRI 6 weeks after inoculation,and appeared in 30% of inoculated animals after 10 weeks. Metastasic focus located brain,chest vertebra, left adrenal gland.Conclusion Metastasis model of highly metastasic human large cell lung cancer cell lines in nude mice has been established.It provides a new metastasis model in nude mice for the study of metastasis of lung cancer.
