Experimental study on properties of xenogeneic deproteinised cancellous bone as scaffold of bone tissue engineering

Objective To study the properties of xenogeneic deproteinised cancellous bone and to prepare scaffold in bone tissue engineering.Methods We made deproteinized bone derived from pigs and goals femoral cancellous bone by physical and chemical treatments. Their morphological features, constitute components and mechanical properties were examined by scanning electron-microscopy,x-rays diffraction analysis,mechanical experimental instrument and so on.Its cellular compatibility was examined by MSCs culture in vitro. Then the scaffolds composited with self mesenchymal stem cells(MSCs) were implanted at intertransverse site of goals.To study the osteogenesis possibility of xenogeneic and heterologous deproteinised bone.Immune response was studied through gross observation.Results It showed that deproteinised cancellous bone maintained natural network pore system.The main organic material is collagenⅠand inorganic is hydroxyapatite.It hardly has any antigen and it has good histocompatibility.Deproteinised cancellous bone composited with MSCs,rhBMP-2 could form cartilage and bone in vivo and more new bone tissue formed gradually as time went by.Conclusion The osteogenesis of deproteinised cancellous bone composited with MSCs in vivo is satisfied.It could be used as scaffold in bone tissue engineering.
