Effect of TGF-β_1 on human kidney tubular epithelial cells′ aptoptosis and cell proliferation dependent of the smad signaling pathway

Objective To examine the effect of TGF-β1 stimulation on smads signaling pathway of human kidney tubular epithelial cells.Methods HKC was stimulated by the recombinant human TGF-β1.The levels of p-smad2/3 and smad2/3 protein were examined by Western blot.At the same time.The level of smad2/3 mRNA was analyzed by reverst transcription polymerase chain reaction(RT-PCR).Flow cytometry was employed to investigate cells′ apoptosis and cycle.MTT assay was used to measure the proliferation of cells at 6,12,24 and 48 hours.Results The protein of p-smad2/3 and smad2/3in HKC obviously increased after TGF-β1 stimulation.TGF-β1 stimulation induced apoptosis of HKC.At the same time,The percent of G0/G1 phase of HKC cell cycle increased apparently.Meanwhile,the proliferation of HKC was markedly inhibited from 6 hour.Conclusion(1)The smad signaling pathway of HKC can be specifically activated by TGF-β1.(2)TGF-β1/Smad signaling patheway may partly cause HKC′s apoptosis and inhibit its proliferation.
