Relationship between expressive variation of APE1 and therapeutic effect by PDT in NSCLC

Objective To explore the relationship between the effect on expressive variation of apurinic/ apyrimidinic endonuclease(APE1) and therapeutic effect by PDT in NSCLC.Methods We analyzed time-effect relationship of expressive variation of APE1 in A549 cells that dealed with PDT by Western blot.Furthermore,to construct Ad5/F35-APE1 siRNA viral interference vector,the knock-down effect of APE1 protein was detected by western blot.Set up control group,Ad5/F35-APE1 siRNA group,PDT group and Ad5/F35-APE1 siRNA+PDT group,and proliferative inhibition was observed by MTT in different groups.Results The expression of APE1 protein that induced by PDT achieved the peak in A549 cells after 24 hours.The APE1 was the most significantly inhibited after 10MOI Ad5/F35-APE1 siRNA recombinant adenovirus infected A549 cells 48 hours.Proliferative inhibition of A549 cells was detected by MTT in Ad5/F35-APE1 siRNA+PDT group that was the most significant,and enhanced with illumination dose.Conclusion Ad5/F35-APE1 siRNA recombinant adenovirus can inhibit efficiently the expression of APE1 that induced by PDT in lung cancer cells,and strengthen lethal effect to lung cancer cells by PDT.
