Extraction and the Physical-chemical Properties of Polysaccharides from Agrimonia pilosa

Crude polysaccharides from Agrimonia pilosa Ledeb were extracted by hot water extraction followed by ethanol precipitation.Orthogonal experiment design L9(34)was applied to optimize the extracting process.The polysaccharides were further deproteined by TCA and purified by chromatography on DEAE-cellulose-52 column.Results showed that the optimal extraction parameters were established as follows: temperature 80℃,extracting time 12 hours,solid-liquid ratio 1∶15.The yield of the polysaccharides was 2.15%.After deprotein,column chromatography purification,main two polysaccharide fractions termed HAPI HAPII were obtained.Partial physicochemical properties were investigated by chemical analysis.The content of protein of HAPI HAPII were 70.08%78.80% respectively.
