Genomic DNA extraction and optimization of RAPD analytic conditions of Michelia wilsonii

The method of genomic DNA extraction and the optimization of RAPD analytic conditions were studied in Michelia wilsonii.The results showed that high grade genomic DNA was obtained by the revised CTAB method.The optimal PCR system for RAPD analysis was as follows: 1.5 mmol/L Mg~(2+),0.8 mmol/L dNTP,240 nmol/L random primer,80 ng DNA template,1.0 U Taq polymerase,in 25 μL reaction system.
