Analysis of DNA Methylation in Silkworm,Bombyx mori Using MSAP Markers

DNA methylation plays an important role in the process of development and gene expression.The extent and pattern of cytosine methylation in 7 silkworm strains were assessed by using the technique of methylation-sensitive amplification polymorphism(MSAP).8.6%~10.9%DNA methylation events were detected in the CCGG sequence of silkworm genomes.Totally 1 156 fragments were amplified with 12 pairs of primers,with 376 DNA methylation polymorphisms.The average proportion of polymorphisms was 32.5%.The results showed that the DNA methylation patterns were different among the silkworm strains.The external cytosine methylation of the CCGG sequence(10.9%) was more frequent than that of internal one(8.6%).
