Preparation and Haemostatic Property Test of a Silkworm Pupa-derived Chitosan Composite Sponge Material

A composite sponge-like haemostatic material was prepared using silkworm pupa-derived chitosan as the major ingredient which was mixed with gelatin and other auxiliary agents by means of freeze vacuum drying.An L16(45) orthogonal experiment was conducted to test the influence of chitosan,gelatin,glutaraldehyde,calcium chloride,and glycerol contents on in vitro blood clotting index(BCI) of the composite haemostatic material.Experimental results showed that the influence of each factor on BCI is in the order of silkworm pupa chitosancalcium chlorideglutaraldehydegelatinglycerol.The optimized mass fraction of each ingredient in the composite haemostatic material was 2.5% silkworm pupa chitosan,3.0% gelatin,0.005% glutaraldehyde,0.1% calcium chloride,and 0.05% glycerol.This composite haemostatic material was further tested on its in vitro haemostatic property and in vivo haemostatic effect using model of rabbit ears with arterial hemorrhage.The results showed that it had good haemostatic effect: its in vitro BCI was extremely significantly lower than that of medical gauze and gelatin sponge;and its haemostatic time and amount of bleeding against arterial hemorrhage of rabbit ears were extremely significantly less than those of medical gauze and gelatin sponge.
