Mulberry Leaf DNJ Extracted Through Microbial Pretreatment and Inhibitory Effect of Extract on β-Glucosidase

1-Deoxynojirimycin(DNJ),an antidiabetic bioactive compound,is abundant in mulberry leaf.In order to improve extraction efficiency of mulberry leaf DNJ,mulberry leaf powder was pretreated with solution of 4 cellulase producing microbial strains respectively,and DNJ was subsequently extracted with diluted hydrochloric acid.When mulberry leaf powder was pretreated by fermentation broth of Trichoderma reesei for 12 h,the highest extraction rate of DNJ reached 1.995 mg/g,being 7.14 times higher than that of the un-pretreated group and 18.20 times higher than that of the distilled water soaking group.Therefore,microbe-assisted extraction method has potential application value in industrial production of mulberry leaf DNJ.Infrared spectroscopy and lignocellulose composition analysis revealed that microbial pretreatment destroyed cell wall of mulberry leaves,resulting in more DNJ released from cells.An enzyme activity assay system composed of cellobiose(substrate),cellulase and mulberry leaf DNJ was established.The results showed that mulberry leaf DNJ had the inhibitory activity to β-glucosidase.
