The Effect of Nitric Oxide on Hepatic Fibrosis in rat

Objective:The purpose of this study was to investigate the role of NO in experimental hepatic fibrosis.Method:Experimental hepatic fibrosis in rats were induced by injecting of thioacetamide in troperitioneally.Rats were divided into control group?TAA group and TAA+L-NAME group.TAA group and TAA+L-NAME group rats were injected TAA(100mg/kg ip) and TAA+L-NAME(200mg/Kg ip).After 13W.Plasma endotoxin content?serum alanine transa minase(ALT)?serum LDH activity and plasma nitric oxide(NO) and hepatic tissue NO?collagen were measured.The changes of hepatic histology were observed by HE and V·G stain.Result:Liver injury and liver fibrosis generated in rats with TAA(ip).The results indicated that ALT?LDH?LPS?collagen and NO contents in plasma and gepatic tissue NO contents were increased significantly in TAA group (vs control group P0.05).those indices were higher in TAA+L-NAME group than those in TAA group.HE and V·G stain show that the grade of hepatic fibrosis and hepatic injury were more serious in TAA+L-NAME group than those in TAA group.Conclusion:Nitric Oxide plays protective role in the process of hepatic fibrosis.
