Optimization of SRAP-PCR program and system in Chinese cabbage

The aim of the research is to establish SRAP-PCR amplification system,which is suitable for Chinese cabbage DNA so as to lay foundation for the construction of gene map and molecular marker in Chinese cabbage.Using Chinese cabbage genome DNA as template,the gradient experiment was conducted on each impact factors in PCR reaction system to screen and establish the optimum system that could amplify high polymorphism,good repeat ability and clear band pattern.The optimum system(total of 25 μL) was as follows: Mg2+ 3.0 mmol·L-1,dNTPs 0.3 mmol·L-1,Taq polymerase 1.5 U,primer 0.2 μmol·L-1,DNA 60 ng.The system could meet the demands for genome SRAP amplification in Chinese cabbage.It is feasible to apply SRAP marker in genetic research in Chinese cabbage.
