Temporal Expression of BpHEN from Betula platyphylla Related to Floral Development by Real-Time PCR

Betula platyphylla inflorescence was sampled in floral phase with temporal interval. Total RNA was isolated from the samples of every period by the method of CTAB, which was subsequently used to synthesize cDNA based on Switching Mechanism At 5′end of RNA Transcript (SMART) strategy. Primers were designed according to the sequence of BpHEN, a gene related to floral development, and then standard samples were prepared after specific amplification, purification, and dilution with series of grads. Expression of BpHEN was analyzed by the method of real-time PCR. Result showed that BpHEN was more highly expressed in female inflorescence than in male inflorescence, and three peaks corresponding to periods of pollination, ovule development, fruit development were found in female inflorescence.
