Microwave-assisted Extraction and Preparative Purification of Polysaccharides from Pholiota adipose

Response surface methodology was used to optimize the microwave-assisted extraction process of polysaccharides from Pholiota adipose. A mathematical model was designed by Box-Behnken central composite design based on single-factor experiments,using extraction time,microwave power,process time for water bath,and ratio of solvent to raw material as influencing factors. Meanwhile,the effects of protein removal were compared in terms of the extraction rate of polysaccharides by Sevag method and Sevag method combined with neutral protease. The optimal conditions for microwave-assisted extraction of polysaccharides from P. adipose were obtained as extraction time 2min,microwave power 490W,process time for water bath 1.6h,and ratio of solvent to raw material 1g∶21mL. The yield of polysaccharides could reach 17.694% at the optimal conditions. The purity of polysaccharide for the combined method was 72.88%,with an increase of 44.71% compared with Sevag method.
