Impacts of Anisodamine(654-2) Combined with Exogenous Pulmonary Surfactant on Tomor Necrosis Factor (TNF-α),Structure Changes of Lung Tissue During Pig's Lung Ischemic-reperfusion Injury

Objective To study the effect of anisodamine (654-2) combined with exogenous pulmonary surfactant (PS) on tumor necrosis (TNF- α) ,structure of lung tissue changes during pig's lung ischemic-reperfusion injury. Methods 24 pigs ,approximately 45-day-old, were randomly divided into four groups: a control group,a 654-2 group,a PS group and a 654-2 plus PS group. The TNF-α concentration of every lung was analysed with ELISA methods at the point of pre-clamping right pulmonary aortery,post-clamping 60',90'(opening),post-unclamping 60',90',120'.Finally ,we examined the pathologic changes of the lungs. Results After clamping right pulmonary artery , TNF-α concentrations in every experimental groups were significantly lower than that of the control group (all P0.01), TNF-α concentration of 654-2+PS group was significantly lower than that of the other experimental groups (P0.01), There was significant alveolar edema, leukocyte infiltration and bleeding in the lung of the control group .The histological examination proved that the degree of pulmonary injury was significantly less in the experimental groups ,especially in the 654-2+PS group. Conclusion 654-2 and exogenous PS have a protective effect on pulmonary function during ischemic-reperfusion injury .Combinnative treatment of 654-2 plus PS can achieve a better protetive effect than that of 654-2 or PS alone.
