Study on differentially expressed genes in laryngeal squamous cell carcinoma by cDNA microarray

Objective: To screen the differentially expressed genes between laryngeal squamous cell carcinoma and normal laryngeal tissue using cDNA microarray. Methods: The PCR products of 4 096 genes were spotted onto a chemical material coated glass plates in array. DNAs were fixed onto the glass plate after series of treatments. The total RNAs were isolated from the tissues, and were purified to mRNAs by Oligotex. Both mRNAs from the laryngeal squamous cell carcinoma and the normal tissue were reversely transcribed to cDNAs with the incorporation of fluorescent dUTP to prepare the hybridization probes. The mixed probes were hybridized to the cDNA microarray. After high stringent washing, the cDNA microarray was scanned for the fluorescent signals and showed differences between 2 tissues. Results: Among the 4 096 target genes, 211 356(5.15% 8.87%)genes whose expression level differed in 2 cases and 36 (0.88%) genes differed in 3 cases between the normal and carcinoma tissues in all 4 cases. Conclusion: cDNA microarray technique is effective in screening the differentially expressed genes between 2 different kinds of tissue. Further analysis of those obtained genes will be helpful to understand the molecular mechanism of malignant carcinoma. [
