Effect of sustained compressive stress on content of IGF-Ⅰ and osteocalcin at experimental fracture sites in rabbits

Objective:To explore the effect of sustained compressive stress on the contents of IGF-Ⅰand osteocalcin at experimental fracture sites fixed by swan-like memory connector(SMC) in rabbits.Methods:Totally 30 rabbits were used to establish experimental fracture model.An osteotomy was performed on one humerus in midshaft by wire saw at random,and the bone fracture was stabilized by SMC(SMC group).The contralateral humerus was osteotomized in the same way and was fixed with 4-hole dynamic compress plate(DCP) as control group(DCP group).Five rabbits were sacrificed each time and bone samples were excised at 2,3,4,6,8 and 12 weeks postoperatively.The contents of IGF-Ⅰand osteocalcin at fracture sites were measured with radioimmunoassay.Results:The contents of IGF-Ⅰand osteocalcin increased at the subsequent time periods in both groups.At 3,4 and 6 weeks after operation,the IGF-Ⅰcontents in SMC group [(2.98±0.32) ng/mg,(3.36±0.18) ng/mg,and (3.47±0.17) ng/mg,respectively] were significantly higher than that in DCP group [(2.54±0.21) ng/mg,(2.78±0.43) ng/mg,and (3.06±0.62) ng/mg,respectively](P0.05).At 4,6 and 8 weeks postoperatively,the osteocalcin contents in SMC group [(387±23) ng/mg,(395±31) ng/mg,and (416±37) ng/mg,respectively] were significantly higher than that in DCP group [(323±16) ng/mg,(356±51) ng/mg,and (378±26) ng/mg,respectively](P0.05).Conclusion:The sustained compressive stress produced by memory fixation of SMC promotes the excretion of IGF-Ⅰand osteocalcin at fracture sites in rabbits.[
