Preparation of oral insulin sustained-release microspheres

Insulin-loaded polyacrylate sustained-release microspheres were prepared using liquid process. The effects of polymer concentration and agitation rates on the size of microspheres and drug incorporation efficiency were studied by scanning electronic microscopy (SEM) and UV-spectrophotometry. The results show that the mean diameter of microspheres decreases with the increase of stirring rate and the decrease of polymer concentration, meanwhile the drug loading efficiency increases with the increase of stirring rate. Besides, the content of emulsifier, temperature and drug payloads all have certain effects on microspheres formation. Finally, the content of insulin in the microspheres was determined.
