Development of gold immunochromatographic assay(GICA) for rapid detection of Salmonella typhimurium

Objective To develope a rapid and simple new method to detect Salmonella typhimurium.Methods Gold immunochromatographic assay(GICA) was established to detecting Salmonella typhimurium by combing colloidal gold technique with immunochromatographic assay.Besides its sensitivity,specificity and stability were studied.Results The optimum marked amount of IgG(rabbit anti mouse) of one-milliliter colloidal gold was 24μg,the optimum concentration of the coating Ab was 1mg·ml-1.The NC film with the best absorbing capability for protein was selected from Millipore company,which diameter was 0.45μm.The prescribed minimum of GICA test strip to detect Salmonella typhimurium was 105cfu·ml-1,which attained the techenical index.It took only 5~10 min from detecting to judging the result.GICA test strip could be stored at 4℃ and room temperature for half a year,which showed the same result.Conclusion GICA detecting Salmonella typhimurium is rapid,sensitive,specific and stable.It is particularly fit for routine test of country sanitary unit.
