Production of transgenic millet plants via particle bombardment

DONG Yun zhou 1,DUAN Sheng jun 2\j(1.Biotechnology Research Center,Chinese Academy of Agricultural Sciences,Beijing 100081,China;2.Institute of Millet Crops,Hebei Academy of Agricultural Sciences,Shijiazhuang 050031,China)  
Inflorescence of millet crop,Yugu No.2,was bombarded by JQ 700 particle gun.The plasmid of pBI121,which containing GUS reporter gene and npt Ⅱ selective marker gene,was used in this study.Totally 908 kanamycin resistance (Kan R) calli was produced in selective medium with 150 mg/L Kanamycin.Five of Kan R calli were characterized out with green shoots which differenciated 16 green plantlets.GUS histochemical assay demonstrated that three of them is positive.Southern blot confirmed that one plant is transgenic.The transgenic frequency is 0.05%.
