Establishment of the Semi-quantitative RT-PCR System of Sulphur Transporter Genes in Wheat

With α-Tubulin gene(Ta) as the inner control,the total RNA of wheat roots was extracted and reversely transcribed to form cDNA to further study the expressions of sulphur transporter genes in wheat suffering drought stress;The fragments of the two genes were PCR amplified at the same cycle numbers at the exponential phase of ST and Ta in different tubes.Mg~(2+) concentration,annealing temperature and cycle number of PCR system were optimized analyzed in their repeatability and precisions,and then a stable and convenient semi-quantitative RT-PCR system was established.This system can be used in comparing the expressions of sulphur transporter genes in different wheat materials and its two pairs of primers are capable of stabilizing the RT-PCR system because both of them are intron-spanned;In addition,the system provides a reference frame to establishing the reverse transcription system of wheat mRNA.
