In Vitro Culture and Regeneration System of Pepper(Capsicum annuum)

Nine pepper varieties(Capsicum annuum) were used to investigate the effects of such factors as different hormone combinations,genotypes,explant types and seedling ages and AgNO_3 on adventitious bud differentiation and elongation of pepper explants.It was shown that the 6-BA/IAA ratio was conducive to the differentiation and regeneration of pepper explants at 3∶1 and unsuitable for the elongation of the regenerative buds;different pepper varieties differed greatly in their regeneration capacities which ranged within 13.3%~90.0%.In pepper,the cotyledons had stronger regenerative capacity than the hypocotyls and thus were good explants;the explants from 12~16 day old seedlings had a high differentiation rate;the addition of AgNO_3 at 4 mg·L~(-1) could raise the average differentiation rate by 16.9%.Comparatively speaking,the optimal medium for the differentiation of pepper buds was MB_5+5 mg·L~(-1) 6-BA+0.5 mg·L~(-1) IAA+4 mg·L~(-1)AgNO_3,the medium for the bud elongation was MB_5+3 mg·L~(-1) 6-BA+1.0 mg·L~(-1) IAA+2 mg·L~(-1) GA_3+4 mg·L~(-1) AgNO_3 and the medium for rooting culture was 1/2 MS+0.2 mg·L~(-1) IAA+0.1 mg·L~(-1)NAA.
