CBF1 Gene Transgenic Strawberry and Increase Freezing Tolerance

To improve strawberry(Fragaria ananassa Duch.) freezing tolerance,cultivars "Honeoye" was studied by genetic transformation.An efficient method of genetic transformation of strawberry was leaf disc by Agrobacterium-mediated method.Transformants were obtained using a binary vector plasmid pBPCBF1 which contains a cauliflower mosaic virus 35S(CaMV35S) promoter driven CBF1(C-repeat binding factor) gene.Adventitious shoots were induced on MS+3.0 mg/L 6-BA+0.1 mg/L 2,4-D medium supplemented with 25 mg/L kanamycin.Results of molecular analysis by PCR and Southern blots confirmed the integration of CBF1 gene into the genome of the strawberry.Electrolyte leakage tests showed that transgenic plants of electrolyte leakage ratio were lower than control's.Plant survival tests demonstrated that 65% of transgenic plants were wilting and 91% of control plants were wilting under-2~0℃ for 7 days,74% of transgenic plants were survival and 18% of control plants were survival under 22~25℃.The results showed that expression of CBF1 gene increased the freezing tolerance of transgenic strawberry.
