Embryosac Development of 'Tosa-Buntan' Pummelo [Citrus grandis(L.) Osb.] and Generational Chromosome in Somatic Cells

Selecting flower buds of 'Tosa-Buntan' pummelo Citrus grandis in different periods and young leaf with 3～5 mm in length of F1 hybrid filial generations were used to observe the ovary structure of female and determine the progenies,ploidy of F1 hybrid filial generations with paraffin section technology and method of chromosome flaking for the citrus plant.The results showed that:(1) the megasporogenesis and the embryosac development characteristic have been tracked.Clear and exact chromosome images of F1 hybrid filial generations were also obtained.The observing result of female parent with paraffin section provided the basement for further confirming the 2n female gamete's best induction period.(2)The observing result of somatic cell chromosomes of F1 hybrid filial generations indicated that both normal diploid and triploid are existed in hybrid filial generations and offers the precondition for further affirmance of the types and possible genetic effects of 2n gamete with in situ hybridization.
