CHANGES IN THE CALMODULIN LEVEL AND EFFECTS OF GA_3 AND ABA ON CALMODULIN LEVEL DURING THE GERMINATION OF CORN SEEDS

Using antibody against native wheat calmodulin,a competitive indirect enzvme-linkedimmunosorbent assay(ELISA)was developed for the quantitation of calmodulin in plants.The linear range of calibration ctirve was between 2 and 1400 ng calmodulin.The meanCV% of ELISA in intra-assay and inter-assay were 7.9% and 10.6% respectively.The levelof calmodulin in germinating corn seed embryos were measured by ELISA.During the ger-mingation process,calmodulin content of per fresh weight and per mg protein of embryos in-creased.The increase of calmodulin is promoted correspondingly with germination promotionby gibberellic acid(GA_3);and the increase of calmodulin is blocked by abscisic acid(ABA),which simultaneously also blocks germination.These results suggest that calmodulin couldplay an important role in the regulation of germination of corn seeds.
