AANAT gene-engineered embryonic stem cells decrease the amyloid β secretion of APP-overexpressed PC12 cells

Objective To construct the arylalkylamin N acetyltransferase (AANAT) gene engineered ES cells and study its possible effects on therapy of Alzheimer's disease. Methods Neomycin gene was transfected into human embryonic fibroblasts (HEF) by liposome mediated method. Using this transfected HEF as feeder layer, ES cells were transfected with AANAT gene. The conditioned medium collected from AANAT gene transfected ES cells was used to culture APP overexpressed PC12 cells. Amyloid β was detected by radioimmunoassay. Results ①HEF transfected with neomycin gene could keep the undifferentiated condition of ES cells; ②The conditioned medium of AANAT gene engineered ES cells could decrease Aβ secretion. ③Melatonin had the same effect on the decrease of Aβ secretion of APP overexpressed PC12 cells. Conclusion AANAT gene engineered ES cells have been constructed successfully. These cells can obviously decrease Aβ secretion of APP overexpressed PC12 cells and may play an important role in the therapy of AD disease.
