Effects of the energy controllable steep pulses on human ovarian carcinoma cell in vitro

Objective To investigate the effects of energy controllable steep pulses on the biological properties of human ovarian cell in vitro . Methods After action by different intensities of steep pulses, the acute and delayed effects on human ovarian carcinoma cell line SKOV3 were investigated by measuring the cell growth curves and by 3 (4,5 dimethylthiazol z yl) 2,5 dipheny tetrazolium blue (MTT) colorimetric assay. Cell cycle changes were detected by flow cytometry (FCM). Morphological changes were observed by inverted microscope, light microscope and scanning electron microscope. Results MTT assay demonstrated the effects of steep pulses on SKOV3 cells were in a dose dependent manner. The cell growth curve showed that the highest dose mainly resulted in acute effects. Analysis of cell cycle indicated that S and G 2/M phase cells decreased but G 0/G 1 cells increased in a dose dependent manner. The morphological dynamic changes of SKOV3 cells in the pulse fields showed that tumor cells had negative charges, moving to anode. Results of ultrastructure and scanning electron microscope showed that steep pulses could bring about irreversible electrical breakdown (IREB) of cell, leading to necrosis of tumor cells. In addition, the function of electrolysis by steep pulse changed the normal pH of the living circumstances of tumor cells and consequently, strong acid and alkali ulteriorly induced the death of cells. Conclusion Energy controllable steep pulses can change not only the features of human ovarian carcinoma cell by acute and delayed effects, but also the normal living circumstances by means of the effects of electrolysis, which, finally, results in death of tumor cells.
