Effects of cholesterol inhibitor lovastatin on the expression of caspase-3 in HL-60 cells

Objective To study the effects of lovastatin (LOV) on the expression of caspase-3 in HL-60 cells and the possible molecular mechanisms of proliferation and apoptosis in HL-60 cells. Methods After treatment with LOV at the dosages of 4, 8, 16 μmol/L for 1-4 d, the cellular proliferation capacity was observed by MTT and growth curve assay. The cell cycle distribution and the apoptotic rate were measured by flow cytometry (FCM). The cell apoptosis was measured by DNA ladder assay. The expression of caspase-3 mRNA was analyzed by RT-PCR. The protein expression of caspase-3 was analyzed by Western blotting. Results Lovastatin could inhibit the proliferation of HL-60 cells and arrest HL-60 cells in the G 0/G 1 phase of cell cycle, resulting in increased apoptotic rate and appearance of the DNA ladders in LOV-treated cells. The protein expression of caspase-3 was up-regulated in HL-60 cells treated with LOV, but no obvious change of the expression of caspase-3 mRNA was found. The effects of LOV were in dose- and time-dependent manners. Conclusion Lovastatin could inhibit cell growth, induce apoptosis, and up-regulate the protein expression of caspase-3, which may play an important role in cell apoptosis induced by LOV.
