Antioxidation of mythic fungus broth on senile rat models induced by D-galactose

Objective To study the effect of mythic fungus broth (FMF) on the free radicals, erythrocyte membrane, sexual function and ultrastructure of the brain mantle in D-galactose induced senile rats. Methods ①Thirty SD rats of 24 months old were divided into 3 groups: water control, FMF group (1.2 g/kg) and vitamin E group (0.8 g/kg). The action of antiaging in rats was studied by determining the oxidative activity of the plasma, the content of vitamin C, glutathin and malondialdehyde in the blood serum and the brain tissue. ②Sixty SD rats of 6-7 months were divided into 6 groups: control group, model group, 0.3 g/kg FMF group, 0.6 g/kg FMF group, 1.2 g/kg FMF group and vitamin E group. All rats except the control group were injected subcutanously with 1% D-galactose at the dose of 50 mg·kg~(-1)·d~(-1). The antiaging action was studied by measuring the activities or content of superoxide dismutase (SOD), malondialdehyde (MDA), glutathione peroxide (GSH-Px), monoamine (MAO) and lipofuscin (LPF) in the blood serum or/and the brain tissue. The protein content in the brain tissue was determined by Coomassive technique. The ultrastructural change in the cerebral neurons was observed by transmission electron microscope.③The effect of FMF on the erythrocyte membrance was studied by assaying the MDA content. ④The change of sexual function was studied by the copulation test in 40 female and 40 male SD rats of 4-5 months. Results ①In the aging rats, the 1.2 g/kg FMF increased the oxidative activities of the plasma (P0.01) and the content of vitamin C of the blood serum(P0.01), decreased the MDA content in the blood serum and the brain tissue (P0.05), but had no significant effect on the GSH. ② 0.3 g/kg FMF enchanced the GSH-Px activities (P0.01),decreased the MDA content in the blood and brain tissue (P0.01) and the LPF content in the brain tissue (P0.05), but had no effects on other enzyme and metabolites; 0.6 g/kg FMF decreased the LPF content (P0.05) and MDA content (P0.01), enchanced the SOD activities (P0.05); 1.2 g/kg FMF increased the SOD activities(P0.05) and GSH-Px content (P0.05), decreased the MDA content (P0.01 or 0.05) and LPF content (P0.05), and the MAO activities (P0.05) in the blood serum and/or the brain tissue. D-galactose increased lipofuscin, made mitochondria swollen and degenerated, mitochondria broken and vacuolated, nucleus teratic, nuclear membrane plicated, nucleolus shifted to edge, and Golgi complex vacuolated. The FMF improved remarkedly the changes induced by D-galactose. ③ 0.6 g/kg FMF and 1.2 g/kg FMF decreased the contents of the MDA in the erythrocyte membrane (P0.01), but 0.3 g/kg did not. ④In the test of strengthening sexual function, 1.2 g/kg FMF shortened the latency of the penis erection (P0.01). In the mating test, 0.6 g/kg FMF increased the number of capture and ejaculation (P0.01), shortened the latency of capture (P0.01) and ejaculation (P0.01), 0.3 g/kg FMF only shortened the latency of capture (P0.05). Conclusion FMF can delay aging, decrease lipid oxidation, increase the activity of antioxidase, enhance the sexual function of male rats.
